I'm investigating the rate of reaction between peroxydisulphate(VI) ions and iodide ions. However, I am a little confused with regards to the method of monitoring the rate of reaction using colorimetry.

The most direct way of doing this is to prepare a series of solutions of iodine (in KI) with known concentrations covering the expected range of concentrations produced by the reaction. Measure the absorbance of each solution, using an appropriate filter or wavelength setting, and plot a graph of absorbance against concentration. This is your calibration curve.
 
Now do the reaction in the colorimeter, taking readings of absorbance at measured time intervals. Use the calibration curve to convert these readings into concentrations.
